A high-performance liquid chromatographic method for the determination
of the HIV protease inhibitor saquinavir in human plasma, saliva, and
cerebrospinal fluid is described. Saquinavir was extracted from sampl
es using C-2 extraction columns prior to ion-pair, reversed-phase high
-performance liquid chromatography with ultraviolet detection at 239 n
m. The method has been validated over the range of 2.5-4000 ng/ml usin
g a 0.6-ml sample volume. This assay has been used for the analysis of
saquinavir in plasma and saliva of HIV-1-infected patients. (C) 1997
