6,7-Dimethoxycoumarins having various reactive groups such as bromoace
tyl-, hydrazinocarbonyl- and chlorocarbonyl-groups at 3-position on co
umarin ring were synthesized for the construction of a fluorescent der
ivatizing reagent-kit for carboxylic acids, aldehyde/ketone, alcohol,
phenol and amines. These reagents were evaluated from their reactiviti
es with model compounds and fluorescent characteristics of products ob
tained. Of the reagents, 3-bromoacetyl- and 3-chlorocarbonyl-6,7-dimet
hoxycoumarins were found to afford highly fluorescent products with ab
ove 0.8 in quantum yields for carboxylic acids, alcohol and aliphatic
amines. It was found that the construction of reagent-kit by using a s
ingle fluorophore selected after consideration of its emission mechani
sm can be significant and useful as an approach for the efficient reag
ent development.
