A new anion-exchange HPLC method for separation of gangliosides has be
en developed using TMAE (trimethylaminoethyl)-Fractogel as the station
ary phase and a gradient system of ammonium acetate in methanol for el
ution. Chromatography of mouse brain gangliosides resulted in separati
on according not only to their degree of sialylation into mono-, di-,
tri- and tetrasialogangliosides, but also in separation of positional
isomers within the four elution domains, e.g. G(D1a) (IV(3)Neu5Ac,II(3
)Neu5Ac-GgOse(4)Cer) and G(D1b) (II3(Neu5Ac)(2)-GgOse(4)Cer) of the di
sialoganglioside fraction. Furthermore, base-line separation of alpha
2-3 and alpha 2-6 sialylated neolacto-series gangliosides e.g. IV(3)Ne
u5Ac-nLcOse(4)Cer and IV(6)Neu5Ac-nLcOse(4)Cer, isolated from human gr
anulocytes, was achieved and pure fractions of each ganglioside were o
btained. Thus, the high-resolution power of the strong anion-exchanger
TMAE-Fractogel allowed the isolation of pure gangliosides on a prepar
ative scale by one-step column chromatography.
