An improved method is described for evaluation of the dissociation rat
e constant for the solute-matrix interaction from affinity chromatogra
phic elution profiles in which the flow-rate dependence of the varianc
e reflects the combined effects of chemical and partition kinetics. Re
sults of a previous investigation of the interaction between saccharid
es and concanavalin A immobilized on CPG 3000 are reconsidered in term
s of the new procedure, which takes advantage of an analytical express
ion to separate the two contributions. Although the revised analysis c
onfirms the adequacy of the earlier empirical approach to this problem
, it also renders redundant the necessity for the empiricism.
