Clostridial hydrogenase was introduced into a cyanobacterium, Synechoc
occus elongatus, by direct electroporation. The introduced hydrogenase
activity was 11 nmol H-2 evolved with reduced Methyl Viologen per mg
chlorophyll. The cells with clostridial hydrogenase showed simultaneou
s light-dependent evolution of H-2 and O-2. This method will be applic
able to assess a variety of hydrogenases for hydrogen production coupl
ed with cyanobacterial photosynthesis.
