Ah. Krotz et al., IMPROVED IMPURITY PROFILE OF PHOSPHOROTHIOATE OLIGONUCLEOTIDES THROUGH THE USE OF DIMERIC PHOSPHORAMIDITE SYNTHONS, Nucleosides & nucleotides, 16(7-9), 1997, pp. 1637-1640
Phosphorothioate oligonucleotides synthesized through assembly of dime
ric phosphoramidite synthons show a significantly improved impurity pr
ofile compared to oligomers synthesized through coupling of standard m
onomer phosphoramidites. A greater than 70% reduction of the (n-1)-mer
population and a ca. 50% reduction of phosphodiester linkages has bee
n achieved.
