SEPARATION AND PURIFICATION OF BOVINE POLYMORPHONUCLEAR LEUKOCYTES WITH FICOLL-PAQUE

Studies were conducted to selectively isolate polymorphonuclear leukoc ytes from bovine blood. Following a short, low speed centrifugation a leukocyte rich fraction was collected. This fraction was diluted 1:1 w ith Iscove's Modified Dulbecco's Medium prepared and layered onto Fico ll-Paque and centrifuged. PMNs and erythrocytes migrated through the d ensity gradient and formed a pellet at the bottom of the tube. Erythro cytes were selectively lysed leaving a highly purified PMN preparation . The purity and viability of the resultant PMN fraction makes this te chnique very useful for subsequent immunological assays.
