A HPLC method for rapid analysis by class of the components during oli
ve oil enzyme hydrolysis is presented; Samples for analyses were taken
directly from a reaction mixture without preparation. A single, short
column with stationary phase RP18 and the RI detector were used. Elut
ion was carried out under isocratic conditions with ternary mobile pha
se composed of acetone, acetonitrile and isooctane. In the presence of
remarkable amounts of water the optimum composition of the phase is 5
0:35:15 volume %. The quantitative results were obtained basing on num
erical resolution of the partly overlapped peaks, assuming gaussian sh
ape of the peaks. The analysis being rapid (12-15 min) and simple is s
uitable for kinetic studies of the hydrolysis and/or synthesis of acyl
glycerols with the immobilised enzyme catalyst.
