An overview of recent advances in the surface modification of colloida
l particles to oppose uptake by the mononuclear phagocyte system (MPS)
is presented. First, we describe the colloidal particles and hydrophi
lic coating materials investigated, with particular focus on the liter
ature concerning particles other than liposomes. Then the pharmacokine
tics and biodistribution of these MPS-opposing systems are summarized.
Finally, the mechanism behind the MPS-avoidance phenomenon is discuss
ed in the light of the concept of steric stabilization, We conclude th
at the literature reviewed provides enough promise for anticipating th
erapeutic and diagnostic applications of surface-modified nanoparticle
