An optimum condition for the separation of large DNA molecules from Pl
eurotus cornucopiae (Paulet) Rolland var. citrinopileatus (Sing.) Ohir
a was investigated by using the pulsed-field gel electrophoresis (PFGE
) method. A clear separation of the DNA molecules was achieved by, a c
ombination of switching intervals and running voltages. Nine and seven
bands were separated from the dikaryon and the monokaryon, respective
ly, within 96 h. They ranged in size from 1.7 to 3.7 Mb. Their genome
sizes were estimated as about 55.2 Mb and 27.8 Mb, respectively.
