Hygromycin-resistance (Hy(R))-based Escherichia coli-mycobacterial shu
ttle plasmids have high efficiencies of transformation and a broad myc
obacterial host range. We have introduced a lacZ alpha (encoding the a
lpha-polypeptide fragment of beta-galactosidase (beta Gal))-multiple c
loning site cassette into a Hy(R)-based shuttle vector to generate a p
lasmid with nine unique cloning sites and the added feature of beta Ga
l color selection in appropriate E. coli host strains.
