A scheme for isolation and purification of cellulolytic enzymes from P
enicillium verruculosum is suggested which includes purification of th
e proteins as a group by ammonium sulfate precipitation, ion-exchange
column chromatography on DEAE-Spheron, and chromatofocusing in a pH gr
adient followed by fine enzyme separation by high performance ion-exch
ange column chromatography on Macro prep Q. Five endoglucanases and tw
o cellobiohydrolases which are homogeneous by analytical isoelectric f
ocusing and gel chromatography were isolated. The molecular weights an
d isoelectric points of these enzymes varied from 19,100-114,800 dalto
ns and 3.70-4.38, respectively. The carbohydrate contents are 8.1-48.3
%. The activities of the isolated cellulase complex enzymes toward var
ious cellulose species and substrates with different glycoside bond ty
pes were determined.
