DC-cyclic voltammetry was employed for characterization of an active s
ite of galactose oxidase(GAO) coupled with some electrode-active media
tors and alkaline shift of the pH dependence in electrocatalytic react
ion. We discussed on difference of the reactivity of various mediators
with native and modified GAG. (Ferrocenylmethyl)trimethylammonium per
chlorate gave the highest catalytic reactivity in ferrocene derivative
s used as mediators of GAG. These electrocatalytic reactions were esti
mated to be related with negative charge near the active site of GAG.
Moreover alkaline shift of the electrochemical behavior was caused by
a local increase of proton which was produced during oxidation of a su
bstrate.
