Inclusion of mycological peptone in the base medium of X-gal plates fa
cilitates the detection of isolates of Saccharomyces cerevisiae produc
ing beta-galactosidase due to improved colour development. Substitutio
n of tryptone with mycological peptone in a standard Escherichia coli
medium (LB) also facilitates colour development on X-gal plates and ob
viates the need for an inducer. The addition of mycological peptone to
the base medium of X-gal plates offers a cost-effective method to imp
rove the detection of the beta-galactosidase gene in recombinant strai
ns of S. cerevisiae and E. coli.
