This work was concentrated in the analysis of the PEG (polyethylene gl
ycol) mediated genetic transformation method for rice protoplast. The
optimisation of the transformation efficiency was made by the analysis
of the transient expression of the gusA gene in protoplasts, the meth
od of analysis chosen was the fluorometric assay. The protoplasts were
isolated from cell suspensions of rice. In this work a technical impr
ovement was achieved by optimising the transformation rates for rice p
rotoplast transformation with the PEG method. Some parameters as gradu
al dilution, amount of plasmid DNA, PEG concentration, and protoplast
density proved to be very important for the PEG-mediated transformatio
n efficiency.
