A HIC support has been synthesized by using the domestic silica with m
ajor pore diameter, The mixture of six standard proteins, alpha-Amylas
e in saliva and rIFN-gamma from E. Coli, were separated through gradie
nt elution by high-performance hydrophobic interaction chromatography
respectively. The chromatographic column has less irreversible adsorpt
ion. alpha-Amylase and lysozyme used in this study are almost quantita
tively recovered on this column, The effects of salt concentration in
eluent and temperature on retention behaviours of proteins were invest
igated with packing prepared, and the chromatographic attribution of t
he support has been demonstrated.
