A new inhibitor of acyl-CoA:cholesterol acyltransferase (ACAT), design
ated lateritin, was isolated from the mycelial cake of Gibberella late
ritium IFO 7188 by successive purification procedure of solvent extrac
tion, silica gel column chromatography and reverse phase HPLC. Spectro
scopic analyses of the compound yielded thyl)-3-phenylmethyl-1,4-perhy
drooxazine-2,5-dione as the proposed structure. Lateritin inhibited ra
t liver ACAT activity by 50% at a concentration of 5.7 mu M. This inhi
bition was time-dependent and irreversible.
