A high-performance liquid chromatographic (HPLC) method with fluoresce
nce detection for the determination of vinorelbine in plasma is descri
bed. The technique was derived from that published by Debal for an ass
ay of vinorelbine in cell culture medium. The modifications concern th
e preparation procedure for plasma samples (a two-step liquid-liquid e
xtraction from plasma is described), optimization of the mobile phase
composition, and use of a single C-18 column. These changes resulted i
n an improved sensitivity and reproducibility of the assay and led to
its feasibility for clinical pharmacokinetic studies. The range of the
assay is 2 to 1000 ng/ml.
