The aim of this study was to test a new chromatographic method for the
quantitative determination of metallothionein (MT), in order to deter
mine the level of exposition to heavy metals. For MTs isolation covale
nt affinity chromatography was applied, which is a modern technique of
separation with good repeatability and reproducibility, allowing spec
ific isolation of the thiolproteins. Fundamentals of indirect determin
ation of the contents of metallothionein protein were worked out by es
timation of the quantities of metal bound to metallothionein protein a
nd adsorbed on covalent affinity gel during a separating process.
