An analytical procedure was developed to determine triclosan and its c
onjugates in human plasma. After enzymatic cleavage of the glucuronide
and sulfate conjugates, the analyte was separated from matrix constit
uents by liquid-liquid extraction. The phenolic hydroxy group of tricl
osan was derivatized with acetic anhydride. The resulting ester was ch
romatographed on a 60 m GCLC-column. An ECD was used for detection and
quantitation. The limit of quantitation for triclosan in plasma was a
bout 1 ng/ml. The analytical method was set up to study the pharmacoki
netics of triclosan in healthy volunteers following single and repeate
d administration of different doses of the compound.
