N-2-(p-n-Octylphenyl)-2'-deoxyguanosilne 5'-triphosphate (OctPdGTP) ha
s been synthesized chemically. OctPdGTP inhibited DNA polymerases (pol
) alpha, delta and epsilon from calf thymus, with moderate selectivity
for pol alpha. Mechanistic studies on pol alpha and bacteriophage T4
DNA polymerase revealed competitive and mixed kinetics of OctPdGTP wit
h respect to the substrate dGTP when the enzymes were assayed on activ
ated DNA and oligo dT:poly dA, respectively.
