In an open study the effects of daily i.v. infusions of 60 mu g Prosta
vasin(R) (PGE(1)) for 10-14 days in 12 patients with peripheral arteri
al occlusive disease (stage Db) according to Fontaine on platelet func
tion and some coagulation parameters have been studied. Another 5 pati
ents received some concomitant drug treatment with PGE(1)-infusions. A
slight but nonsignificant inhibition of platelet adhesion was observe
d. Collagen- and ADP-induced aggregation did not change. Platelet indu
ced thrombin generation time (PITT) was not prolonged. A slight nonsig
nificant increase of fibrinogen-levels was recorded during the infusio
n period. During the usual treatment of POAD with 60 mu g PGE(1) daily
for two hours no increased risk of bleeding by inhibition of relevant
platelet functions has to be expected.
