We report an extended whole-mount in situ hybridization procedure for
Drosophila embryos. By using probes labelled with digoxigenin, fluores
cein and biotin, respectively, this protocol allows the detection in t
hree colours of RNAs derived from three different genes. Hybridized pr
obes are detected by consecutive staining with appropriate alkaline ph
osphatase conjugates using different chromogenic substrate combination
s, and serial removal of the antibody conjugates by low pH washes.
