We have isolated human antibody fragments with binding specificities a
gainst the blood group antigens of the ABO and I blood group systems (
B and HI), of the Rh system (D and E) and of the Kell system (Kp(b)),
by selecting a phage-antibody library on human red cells. The fragment
s, expressed in bacteria, were antigen-specific and effective in assay
s including agglutination and immunohistochemistry. Selection of phage
-antibody libraries using intact cells seems to offer a promising alte
rnative to hybridoma technology for the production of antibodies again
st cell surface molecules.
