The frequencies of the A and B alleles of kappa-casein in the 25 indiv
iduals of Bohemian Red strain were detected by the polymerase chain re
action (PCR) and by the restriction fragments length polymorphism (RFL
P). This old strain belongs to the endangered gene reserves. The DNA w
as isolated from the blood. For PCR the primers JK 501 (5'-ATCATITATGG
CCATTCCACCAAAG-3') and JK 302 (5'-GCCCATTTCGCCTTCTCTGTAACAGA-3') were
used, the PCR product was split by the restriction enzyme HindIII. The
AA genotype was found in 6 animals, genotype AB in 14 and BE in 5 ani
mals. The relative frequency of the A allele was 0.52; B allele 0.48;
the level of homozygosity 0.501; relative frequency of the AA genotype
was 0.24; AB 0.55; BE 0.2. The theoretical frequencies of the genotyp
es were not significantly different compared to real ones. From the ci
tations of the relevant literature it is obvious that allele A is prev
alent, mainly in Holstein-Friesian cattle.
