During the last few years, several experiments using off-resonance rf
irradiation have been described for structural and dynamic studies of
biomolecules in liquid state NMR. First, a concise description of the
calculation leading to the expression of relaxation rates and a summar
y of the intrinsic advantages of such methods are given. The resulting
off-resonance ROESY sequence enables the structural study of ail kind
s of molecules. The problem of internal dynamics is then discussed, fo
cussing on the interest of using off-resonance rf fields. Several expe
riments using them are described and compared with other procedures. F
inally the trends of NMR development concerning the study of internal
dynamics are mentioned.
