We performed a focused chromosome analysis on the HL60 cell line by mu
lticolor fluorescence in situ hybridization (FISH), using probes for t
he unequivocal identification of specific chromosome regions and subre
gions. The purpose of this karyotypic re-evaluation rr as to confirm a
nd to characterize in more detail chromosome rearrangements already id
entified by means of classic cytogenetic approaches and recurrently de
tected from the initial establishment of the cell line. The observatio
ns reported may help reassess the potential of the HL60 cell line in u
nderstanding the molecular events underlying the non-random karyotype
alterations associated with acute myeloid leukemias (AML).
