Nucleotide sequences coding for the signal peptides of three secreted
proteins: alkaline phosphatase, outer membrane protein OmpA of Escheri
chia coli, and protein A of Staphylococcus aureus were amplified by po
lymerase chain reaction. Restriction sites for EcoRI and HindIII were
introduced into the 5' and 3' ends of amplified sequences, respectivel
y. Vectors with the signal peptide-coding sequences under the control
of tip and tac promoters were constructed. Thre vectors enable cloning
of genes for foreign proteins at the unique HindIII site. Plasmids fo
r expression and secretion of human growth hormone were obtained.
