The diploid legume species Lotus japonicus was transformed using a dis
armed Agrobacterium strain. By inoculating the cotyledon attachment si
te, we took advantage of the regenerative capability of the plant whic
h spontaneously regenerated transformed shoots. Some of these shoots w
ere propagated as transgenic plants and later transferred to the glass
house to produce seeds. Seeds were germinated and yielded non-chimeric
, transgenic stably modified Lotus plants.
