The conformational flexibility of the linkage mechanism or tether betw
een paramagnetic metal centers is explored to assess optimal structura
l characteristics of multimeric MRI contrast agents. Two dimeric param
agnetic chelates differing in linkage flexibility but similar in symme
try are compared. Relaxivity, water hydration number, and reorientatio
nal times of the chelate groups are obtained, and the results demonstr
ate that greater linkage flexibility reduces overall relaxivity.
