Twenty-three healthy, male volunteers completed this doubleblind, rand
omized, placebo controlled, 2-period crossover study to assess the inf
luence of multiple doses of pantoprazole on single-dose phenytoin phar
macokinetics. During each treatment period, the volunteers received ei
ther one 40 mg pantoprazole tablet or placebo for 7 days. In addition,
a single-dose of 300 mg (3 x 100 mg capsules) phenytoin sodium was ad
ministered on day 4 of each treatment period. A 14-day wash-out period
was allowed between phenytoin administrations. The results indicate t
hat pantoprazole neither affects the rate nor the extent of absorption
, nor the elimination of phenytoin.
