Pg. Penketh et al., SPECTROPHOTOMETRIC ASSAY FOR PROCESSES INVOLVING CHANGES IN HYDROGEN-ION CONCENTRATION IN AQUEOUS-SOLUTION, Analytical biochemistry, 238(1), 1996, pp. 46-49
A strategy for the design of simple colorimetric assays to follow any
process that involves the net generation or consumption of hydrogen io
ns in aqueous solutions over the pH range of 3-10 is described. This p
rocedure relies upon the measurement of the change in absorption when
a weakly or moderately buffered solution of a pH indicator is subjecte
d to a small change in pH. Buffers and indicators are chosen with clos
ely matching pK(a) values. The versatility of this type of assay techn
ique is illustrated using three examples. (C) 1996 Academic Press, Inc
