Protoplast fusion of Lactobacillus plantarum ATCC10012 was examined wi
th an electric field fusion method using its double mutants having dru
g resistance and auxotrophic marker genes. Under the conditions of pul
se width of 150 mu s and an electric field intensity of 6 kV/cm, the f
requency of obtained strains carrying the phenotypes from both mutants
was highest (6.2 x 10(-7)). The frequency was at least one order high
er than the reversion frequencies and that of conjugation. These resul
ts suggest the completion of protoplast fusion.
