This paper describes the development of an electroporation procedure f
or Xanthobacter autotrophicus GJ10. Electroporation has not previously
been described for this species. The procedure was optimized with res
pect to field strength, pulse time and DNA and cell concentration. Hig
h transformation efficiencies of up to 2X10(6) cells transformed per m
u g DNA were found. The conditions optimal for the X. autotrophicus GJ
10 were used to electrotransform 9 other Xanthobacter strains 4 of whi
ch were transformed with high efficiencies. The electrotransformation
procedure described here can be of great help for the genetic availabi
lity of Xanthobacter species.
