An isocratic HPLC system is described which allows the separation of t
he iridoid and indole precursors of terpenoid indole alkaloids, which
are present in a single crude extract. The system consists of a column
of LiChrospher 60 RP select 6 5 mu m, 250 x 4mm (Merck) with an eluen
t of 1 % formic acid-acetonitrile-trichloroacetic acid (100:10:0.25, v
:v:w) at a flow of 1.2 ml/min. In the suspension cultures of Catharant
hus roseus secologanin and tryptophan were detected. In the cultures o
f Tabernaemontana divaricata loganin, tryptophan, and tryptamine accum
ulated.
