Abstract
A procedure for high level tritium labelling at the C2-H position of a denosine 5'-triphosphate ([2-H-3]-ATP, 1), based on the tritiodehaloge nation reaction of 2-bromoadenosine 5'-triphosphate (2) has been elabo rated. This precursor was prepared in a six-step synthesis from guanos ine. The tritiodehalogenation of (2) for three hours over palladium ox ide in phosphate buffer yielded tritium labelled ATP with high specifi c activity, in good chemical yield.
