Recently, it has been discovered that DNA topoisomerases are the targe
t of many anti-cancer drugs which were already widely used in the clin
ic. Using the latest molecular biological techniques much has been lea
rned about the function of DNA topoisomerases in normal cells and in c
ancer cells, and about how anticancer drugs inhibit these enzymes. In
this review we present an overview of the function of human type II DN
A topoisomerases, how they are inhibited by certain drugs and on how c
ancer cells may become resistant to these drugs.
