The estimation of the enzyme kinetic parameters is always a challengin
g problem. This paper covers two methods to estimate the turnover numb
er and illustrates these by the example of laccase enzyme. Both method
s exploit the dynamic behaviour of the enzyme reaction, one of them us
es the derivative values the other the integral ones. The experimental
system is characterized which can be described by the simple Michaeli
s-Menten enzyme reaction scheme. The results of this research are the
estimation of the turnover number, the calculation of the total enzyme
(which is active in this reaction) and the calibration coefficients b
etween absorbance and molar concentration for the product. This calibr
ation is called ''calibration by kinetics''.
