Sixteen withanolides isolated from Iochroma gesnerioides (Kunth) Miers
(Solanaceae) have been assessed for their activities as ecdysteroid a
gonists and antagonists. None of the compounds showed any agonistic ac
tivity, but several showed significant antagonistic activity. With a 2
0-hydroxyecdysone concentration of 5 x 10(-8) M, the ED(50) values for
2,3-dihydro-3 xi-methoxywithaferin A, 2,3-dihydro-3 xi-methoxywithacn
istine, 2,3-dihydro-3 xi-methoxyiochromolide and 2,3-dihydro-3 xi-hydr
oxywithacnistine are 3.5 x 10(-5) M, 1 x 10(-5) M, 5 x 10(-6) M and 2.
5 x 10(-6) M, respectively.
