One of the frequent causes of pollen allergy in our region (Serbia, Yu
goslavia) is the pollen of poplar (Populus canadensis). The aim of thi
s study was to form RAST for the determination of specific anti-Populu
s canadensis IgE antibodies. Affinity purified and radiolabelled (I-12
5) MoAb El was used for forming assay for the determination of specifi
c IgE. By titration of extract of poplar Populus canadensis we determi
ned that the quantity of 0.65 mt extract is needed for coupling of ig
BrCN activated paper discs. Coupling was performed in Na(2)CO3/NaHCO3
buffer pH 10. on 4 degrees C for 48h. Using this newely formed RAST, s
pecific for Populus canadensis we have deteminated anti- Populus canad
ensis IgE antibodies as well. as cross reactivity between pollens of P
opulus canadensis and Populus deltoides.
