Prostaglandin E(1) incorporated in lipid microspheres (lipo-PGE(1)) wa
s evaluated as an inhibitor of ADP-induced thrombus growth and as a th
rombus disaggregating agent following vascular damage in the microcirc
ulation of the hamster cheek pouch. The lipo-preparation was shown to
be significantly more active and longer acting than PGE(1) as an inhib
itor of ADP-induced thrombus formation. In the vascular damage model,
Lipo-PGE(1) was very efficacious as a thrombus-disaggregating agent an
d was significantly more effective when administered after, rather tha
n before, vascular damage and thrombus induction. The data suggested t
hat the lipo-preparation might be targeted to the site of damage.
