D-amino acid oxidase and HRP were separately immobilized on chitosan a
dsorbed on the silica gels to develop a chemiluminescent method, which
was combined with two oxidation reactions of D-amino acid producing H
2O2 catalyzed by D-amino acid oxidase and of Luminol-H2O2 generating C
L catalyzed by HRP together, also was stopped-flow technique used in t
his system. It has a good selectivity and sensitivity. Under optimized
conditions, the detection limit is 1.8 x 10(-8) g/mL, the liner range
of D-amino acid concentration was from 8 x 10(-8) to 1.4 x 10(-6) g/m
L. It was practicable to determine D-amino acid in serum directly with
satisfactory results.
