Abstract
The described staining techniques for glucose oxidase activity on poly acrylamide gels are based on the fact that glucose oxidase reacts with one-electron scavengers in a reaction dependent on pH and pO(2). Gluc ose oxidase separated in PAGE shows reduction of fenicyanide, cuprisul fate, fenicytochrome c and nitrotetrazolium blue reduction leading to sharp coloured bands under aerobic conditions. In contrast to the conv entional peroxidase/o-dianisidine-assay, the staining procedures only need about 30 min and are specific for glucose oxidase using glucose a s electron donor. Enzyme activity can be detected down to 0.2 units.
