A novel stereospecific synthesis of carbocyclic NAD(+) has been develo
ped which allows incorporation of a bisphosphonate linkage in place of
the natural pyrophosphate. This analogue has increased stability to e
nzymatic degradation and has potential as an inhibitor of ADP-ribosyl
cyclase. The synthesis involves two separate Poulter-type couplings to
link the nucleoside moieties together and a Zincke coupling to instal
l the required nicotinamide functionality.
