Experimental design methodology permits to determine experimental cond
itions in order to maximize the enzymatic initial rate reaction. After
determination of factors of the study, we have proposed a model by us
ing a Doehlert uniform shell design. The study of the response equatio
n and the canonical equation has given factor Values that optimize the
experimental response which is the enzymatic initial rate reaction. F
rom the obtained mathematical model, we have thus been able to determi
ne the critical factors as well as Michaelis-Menten constants and Dalz
iel coefficients to specify the kinetic mechanism without supplementar
y manipulations. We have chosen to apply the experimental design by st
udying the kinetics of a well known enzyme: the glutathione peroxidase
.
