RAPID IN-VITRO MULTIPLICATION OF DALBERGIA-SISSOO ROXB

Micropropagation of Dalbergia sissoo Roxb was achieved through in vitr o proliferation of axillary buds from 30 to 40 years old mature tree. Bud-break was achieved within six days when nodal explants were cultur ed on MS medium supplemented with kinetin (9.2 mu m), indole-3-butyric acid (2.46 mu M) and 6-benzyladenine (13.2 mu M). Multiple shoot form ation occurred from nodal explants of in vitro raised shoots on MS med ium with reduced levels of major salts and kinetin. Roots were induced within 5 days on in vitro generated shoots on MS medium supplemented with l-naphthalene acetic acid (0.53 mu M) and indole-3-butyric acid ( 9.8 mu M).
