A new method for the synthesis of N3'-->P5' phosphoramidate oligonucle
otides is demonstrated. This phosphoramidite amine-exchange method gen
erates cycle yields in the 92-95% range, and provides overall yields t
hat are 3-6 times higher than the previously published oxidative phosp
