The pH of the embryonic blood, one of the most important environmental
factors for embryonic cells, was found to range from 8.1 to 8.5 in ch
ick embryos until 108 h after incubation. Based on these results, the
culture medium adjusted to pH 8.0 was used to culture embryonic chick
and quail cells. They were easily subcultured for a long period of tim
e at pH 8.0. This pH culture condition may have wide application for m
anipulating embryonic cells or tissues and establishing cell lines fro
m avian embryos.
