The investigation of the interrelationship between the capacity for ta
rget binding, cytolysis and production of reactive oxygen intermediate
s (ROI) by tumor-associated macrophages (TAM) was carried out, TAM abi
lity to bind and lyse autologous tumor cells in C57BI/6 mice bearing t
ransplanted melanoma B-16 was investigated. The ROI production by TAM
and resident peritoneal macrophages (PM) from intact and tumor bearing
mice was measured in luminol (LM) or lucigenin (LG)-enhanced chemilum
inescent (CL) assay. We showed the low capacity of TAM to bind and lys
e the autologous tumor cells. The amount of CL emitted by TAM in LG-sy
stem was markedly low in comparison with CL of PM from health and espe
cially tumor bearing mice.
