Abstract
The site-specific endonuclease R . AspMI was isolated from Acinerobact er species (strain M) and purified to functional purity. The enzyme re cognizes the symmetrical DNA sequence 5'-AGG down arrow CCT-3' and cle aves it as shown by the arrow, forming blunt DNA ends. The enzyme is a n isoschizomer of endonuclease StuI. Cleavage of the DNA site is inhib ited by dcm-methylation. AspMI is in solution as an similar to 30-kD m onomer.
